A polystyrene-based support for solid phase peptide synthesis (SPPS) was first used by Merrifield in the early 1960s. His article Solid Phase Peptide Synthesis. I. The Synthesis of a Tetrapeptide revolutionized the peptide synthesis industry by introducing the method of solid phase synthesis, which largely increased the efficiency of peptide synthesis.

With the advent of solid-phase techniques, peptide synthesis became increasingly popular and practical in today’s scientific research. One of the main advantages of solid phase peptide synthesis is that a large excess of soluble reagents can be employed to drive reactions to completion; further, the excess of soluble reagents can also be removed through filtration without manipulative losses, which can largely free up researchers from repetitive and tedious workup that are often associated with organic synthesis in solution. Because general and high-yielding reactions can be performed on solid support, solid-phase synthesis is particularly well suited to library generation involving multi-step transformations1.

However, how to choose the right resin for your solid phase peptide synthesis can be tricky as different resin choices can greatly change the synthesis outcome.

The first factor you should consider is the purpose of this synthesis, which plays a crucial role in this resin selection game as it decides or at least indicates whether C-terminal acid or amide is required given the broad application SPPS has. Taking Fmoc-based solid phase peptide synthesis as an example, c-terminal acid and amides are the major two SPPS resin categories. Rink Amide Resin is the most commonly used SPPS resin in the amide family. As for peptide acids, Wang resin is the most commonly used SPPS resin due to the high-yields and purity it can achieve. Despite its popularity, people prefer to buy preloaded resins with fmoc-amino acid attached to wang resins. The reason behind this is that the esterification of Wang resins with fmoc amino acids can be challenging chemically.
